Investigating the basis for the antidepressant effects of Gleditsiae spina using an integrated metabolomic strategy

Overall design

The aim of this study was to investigate the role of taxifolin as a component of Gleditsiae spina in mediating the antidepressant effects of Gleditsiae spina. Depression models in vivo were established using corticosterone. C57BL/6 mice were divided into the following 4 groups (n = 8 in each group): (1) control group, (2) corticosterone group, (3) corticosterone + fluoxetine group (corticosterone plus fluoxetine treatment at the dose of 5 mg/kg), (4) corticosterone + Gleditsiae spina group (corticosterone plus Gleditsiae spina treatment at the dose of 3.33 g/kg), and (5) corticosterone + taxifolin group (corticosterone plus taxifolin treatment at the dose of 0.1 g/kg). The corticosterone injection lasted for 3 weeks, and drugs were administered orally during the model building period. A simultaneous metabolomic approach was used to compare the systemic effects of Gleditsiae spina and taxifolin in urine samples through metabolic networks.
