Title | Integrated network pharmacology and metabolomics to dissect the combination mechanisms of Bupleurum chinense DC-Paeonia lactiflora Pall herb pair for treating depression |
Overall design | In this study, the combined effect of Chaihu-Baishao was evaluated by the chronic unpredictable mild stress (CUMS) rat model. Sprague-Dawley rats were divided into the following 5 groups (n = 8 in each group): (1) control group, (2) CUMS group, (3) CUMS + low dose of Baishao group (stressor plus Baishao treatment at the dose of 7.5 g/kg), (4) CUMS + low dose of Chaihu group (stressor plus Chaihu treatment at the dose of 7.5 g/kg), and (5) CUMS + low dose of Chaihu-Baishao group (stressor plus Chaihu-Baishao treatment at the dose of 7.5 g/kg). The CUMS stress procedure lasted for 4 weeks, and drugs were administered via intragastric once per day during the model building period. Plasma metabolomics based on ultra-high performance liquid chromatography combined with quadrupole time-of-flight mass spectrometry (UPLC-QTOF/MS) was performed to discover the collaborative effect on metabolite regulation. |
