Title | Metabolomic profiling relates tianeptine effectiveness with hippocampal GABA, myo-inositol, cholesterol, and fatty acid metabolism restoration in socially isolated rats |
Overall design | In this study, the comprehensive two-dimensional gas chromatography-mass spectrometry (GC x GC-MS)-based comparative non-targeted analysis was employed on hippocampus of rats to examine the effects of chronic social isolation (6 weeks) and/or chronic tianeptine treatment (10 mg/kg per day) in controls and chronic social isolation to obtain real-time metabolomic fingerprints representative of the underlying molecular changes. Wistar rats were divided into the following 5 groups: (1) control group (no stress, with vehicle treatment), (2) tianeptine group (no stress, with tianeptine treatment at the dose of 10 mg/kg), (3) chronic social isolation group (social isolation, with vehicle treatment), (4) chronic social isolation + tianeptine group (social isolation plus tianeptine treatment at the dose of 10 mg/kg, rats responding effectively to tianeptine treatment), and (5) chronic social isolation + tianeptine resilient group (social isolation plus tianeptine treatment at the dose of 10 mg/kg, rats responding ineffectively to tianeptine treatment). The chronic social isolation procedure lasted for 6 weeks, and tianeptine was administered with intraperitoneal injections during the model building period. Hippocampal and serum samples were used for metabonomic analysis (n =5-6/ group). |
