Title

Metabolomics based on peripheral blood mononuclear cells to dissect the mechanisms of Chaigui granules for treating depression

Overall design

In this study, the chronic unpredictable mild stress (CUMS) rat model was used to evaluate the antidepressant effect of Chaigui granules. Sprague-Dawley rats were divided into the following 4 groups (n = 8 in each group): (1) control group, (2) CUMS group, (3) CUMS + middle dose of Chaigui granules group (stressor plus Chaigui granules treatment at the dose of 8.3 g/kg), and (4) CUMS + high dose of Chaigui granules group (stressor plus Chaigui granules treatment at the dose of 16.6 g/kg). The CUMS stress procedure lasted for 4 weeks, and drugs were administered via intragastric once per day during the model building period. Peripheral blood mononuclear cell (PBMC) metabolomics was used to search the key metabolites and metabolic pathways regulated by Chaigui granules. Six samples per group were used for metabonomic analysis.
