Overall design | In this study, a comprehensive metabolomic analysis was used to explore for the potential mechanism of Baihe-Dihuang Tang in chronic unpredictable mild stress (CUMS)-induced depression in rats. Sprague-Dawley rats were divided into the following 4 groups (n = 7 in each group): (1) control group, (2) CUMS group, (3) CUMS + Baihe-Dihuang Tang group (stressor plus Baihe-Dihuang Tang treatment at the dose of 15 g/kg), and (4) CUMS + fluoxetine group (stressor plus fluoxetine treatment at the dose of 3.6 mg/kg). The CUMS stress procedure lasted for 6 weeks, and drugs were administered via intragastric once per day from the 22 day of the model building period. Untargeted metabolomics based on ultra-high performance liquid chromatography with quadrupole time-of-flight mass spectrometry was performed to detect variations in the metabolites/metabolite categories in the brain, plasma, and urine of rats. The contents of dopamine, norepinephrine, 5-hydroxytryptamine, L-glutamate, xanthine, and adenine in the brain were quantified by targeted ultra performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) metabolomics. |
