Analysis of antidepressant activity of Huang-Lian Jie-Du Decoction through network pharmacology and metabolomics

Overall design

The aim of this study was to explore antidepressant activity of Huang-Lian Jie-Du Decoction (HLJDD), and evaluating active components and potential depression-associated targets. HLJDD was administered on chronic unpredictable mild stress-induced (CUMS) depressive mice. C57BL/6 mice were divided into the following 3 groups (n = 10 in each group): (1) control group, (2) CUMS group, and (3) CUMS + HLJDD group (stressor plus HLJDD treatment at the dose of 590 mg/kg). The CUMS stress procedure lasted for 5 weeks, and drugs were administered via intragastric once per day during the last week of the model building period. Neurotransmitters including c-aminobutyric acid, glutamic acid, 5-hydroxytryptamine, dopamine and acetylcholine in the hippocampus, cortex, striatum and amygdala were determined using LC-MS/MS.
