Title

Determination and application of nineteen monoamines in the gut microbiota targeting phenylalanine, tryptophan, and glutamic acid metabolic pathways

Overall design

In this study, the authors established an underivatized liquid chromatography-tandem mass spectrometry (LC-MS/MS) method for the quantification of nineteen monoamine neurotransmitters and related metabolites in the gut microbiota. The neurotransmitters and related metabolites included phenylalanine, tyrosine, L-dopa, dopamine, 3-methoxytyramine, tryptophan, hydroxytryptophan, 5-hydroxytryptamine, 5-hydroxyindole-3-acetic acid, kynurenine, kynurenic acid, melatonin, tryptamine, indole-3-lactic acid, indole-3-acetic acid, indolyl-3-propionic acid, glutamic acid, gamma-aminobutyric acid, and acetylcholine. This method was validated in fresh fecal samples from chronic unpredictable stress (CUS) model of depression. Sprague-Dawley rats were divided into the following 2 groups (n = 6 in each group): (1) control group, and (2) CUS group. The CUS stress procedure lasted for 8 weeks.
