Title | LC-MS/MS-based quantification of tryptophan metabolites and neurotransmitters in the serum and brain of mice |
Overall design | In this study, a simple, rapid, sensitive and specific liquid chromatography-tandem mass spectrometry method was developed for simultaneous determination of tryptophan, kynurenine, kynurenic acid, 3-hydroxykynurenine, 5-hydroxytryptamine, 5-hydroxyindoleacetic acid, norepinephrine, L-glutamic acid, GABA and acetylcholine in mice serum and the brain tissues in a single chromatographic run. In order to apply this method, the biological samples in normal control and chronic mild stress (CMS) mice were analyzed. BALB/c mice were divided into the following 2 groups: (1) control group, and (2) CMS group. The CMS stress procedure lasted for 6 weeks. Seven to nine samples per group were used for metabonomic analysis. |
