Title

The anti-depression effect of Angelicae Sinensis Radix is related to the pharmacological activity of modulating the hematological anomalies

Overall design

The aim of this study was to elucidate the potential anti-depression mechanisms of Angelicae Sinensis Radix (AS) that are related to the activity of modulating the blood system. The model of chronic unpredictable mild stress (CUMS) was performed to assess the anti-depression effect of AS. A metabolomic approach based on UPLC-MS/MS combined with 1H NMR was employed to analyze the endogenous metabolites and metabolic pathways regulated by AS in depression. Sprague-Dawley rats were divided into the following 5 groups (n = 12 in each group): (1) control group, (2) CUMS group, (3) CUMS + venlafaxine group (stressor plus venlafaxine treatment at the dose of 35 mg/kg), (4) CUMS + low dose of AS group (stressor plus AS treatment at the dose of 7.5g herb/kg), and (5) CUMS + high dose of AS group (stressor plus AS treatment at the dose of 15g herb/kg). The CUMS stress procedure lasted for 4 weeks, and drugs were administered via intragastric once per day during the model building period.
