Identification of proline, 1-pyrroline-5-carboxylate and glutamic acid as biomarkers of depression reflecting brain metabolism using carboxylomics, a new metabolomics method

Overall design

In this study, a sensitive 5-(diisopropylamino) amylamine derivatization coupled with UHPLC-Q-TOF/MS was used to profile carboxylic acids in serum samples of patients with depression. A total of 130 serum samples, including 65 patients with depression (depression group) and 65 healthy controls (control group) were recruited. The training cohort included 19 patients with depression and 15 healthy controls, and the validation cohort included 46 patients with depression and 50 healthy controls. Metabolic profiling was performed based on the samples from the training cohort, then potential biomarkers were validated in the validation cohort. The difference with P < 0.05 was considered as statistical significance.
