Title | GC-MS-based metabolomic study on the antidepressant-like effects of diterpene ginkgolides in mouse hippocampus |
Overall design | Here, we investigated the antidepressant-like effects of diterpene ginkgolides (DG), an important class of constituents in Ginkgo biloba extract (GBE), and studied their effects in the mouse hippocampus using a GC-MS-based metabolomics approach. Mice were randomly divided into the following 5 groups: (1) control group (n = 9), (2) low dose of DG group (DGMI diluted in 0.9% NaCl solution to a dose of 4.06 mg/kg, n = 10), (3) medium dose of DG group (DGMI diluted in 0.9% NaCl solution to a dose of 12.18 mg/kg, n = 11), (4) high dose of DG group (DGMI diluted in 0.9% NaCl solution to a dose of 36.54 mg/kg, n = 11), and (5) venlafaxine group (venlafaxine diluted in 0.9% NaCl solution to a dose of 16 mg/kg, n = 11). Mice were injected intraperitoneally daily between 8:00 AM and 10:00 AM with the same amount of solution for 14 days. Behavioral testing (sucrose preference and tail suspension tests) showed the medium-dose group showed a better antidepressant-like effect, suggesting that the DG antidepressant-like effect was dose related. Samples from control group, medium dose of DG group, and venlafaxine group were used for metabolomics analysis. |
