Title | [Based on plasma metabonomics and network analysis to research the mechanisms of Chaigui granules for treating depression] |
Overall design | The aim of this study was to systematically analyze the antidepressant mechanism of Chaigui granules from the perspective of biological metabolic network in the chronic unpredictable mild stress (CUMS) model of depression. Sprague-Dawley rats were divided into the following 4 groups (n = 8 in each group): (1) control group, (2) CUMS group, (3) CUMS + middle dose of Chaigui granules group (stressor plus Chaigui granules treatment at the dose of 8.3 g/kg), and (4) CUMS + high dose of Chaigui granules group (stressor plus Chaigui granules treatment at the dose of 16.6 g/kg). The CUMS stress procedure lasted for 4 weeks, and drugs were administered via intragastric once per day during the model building period. LC-MS-based plasma metabolomics was used to identify the key metabolites and analyze metabolic pathways. Six samples per group were used for metabonomic analysis. |
