Synergistic effect of several neurotransmitters in PFC-NAc-VTA neural circuit for the antidepression effect of Shuganheweitang in a chronic unpredictable mild stress model

Overall design

The aim of this study was to verify the efficacy by modern methods and investigate the material basis for the efficacy of Shuganheweitang. Different fractions have been prepared and several neurotransmitters in the PFC-NAc-VTA neural circuit that maybe related to the mechanism of chronic unpredictable mild stress (CUMS)-induced depression have been detected. Sprague-Dawley rats were divided into the following 10 groups (n = 10 in each group): (1) control group, (2) CUMS group, (3) CUMS + fluoxetine group (stressor plus fluoxetine treatment at the dose of 1.58 mg/kg), (4) CUMS + low dose of Shuganheweitang group (stressor plus Shuganheweitang treatment at the dose of 3.67 g/kg), (5) CUMS + high dose of Shuganheweitang group (stressor plus Shuganheweitang treatment at the dose of 14.68 g/kg), (6) CUMS + low dose of volatile oil group (stressor plus volatile oil treatment at the dose of 0.018 g/kg), (7) CUMS + high dose of volatile oil group (stressor plus volatile oil treatment at the dose of 0.073 g/kg), (8) CUMS + low dose of polysaccharide group (stressor plus polysaccharide treatment at the dose of 0.56 g/kg), (9) CUMS + high dose of terpenoid group (stressor plus terpenoid treatment at the dose of 0.012 g/kg), and (10) CUMS + high dose of phenolic acid group (stressor plus phenolic acid treatment at the dose of 0.13 g/kg). The CUMS stress procedure lasted for 3 weeks, and drugs were administered via intragastric once per day for 3 weeks after stress. Amino acid (GABA, Glu) and monoamine neurotransmitters (DA, 5-HT, NA, 5-HIAA) in the PFC-NAc-VTA neuro circuit were investigated by UPLC-MS/MS (n = 8 per group).
