Title

5-Fluorouracil induced dysregulation of the microbiome-gut-brain axis manifesting as depressive like behaviors in rats

Overall design

In this study, behavioral experiments were initially performed to determine whether the rats developed depression-like behaviors after 4 days 5-fluorouracil treatment. Sprague-Dawley rats were divided into the following 4 groups: (1) control group (vehicle injection, n = 10), (2) 5-fluorouracil group (5-fluorouracil intraperitoneal injection of 40 mg/kg once a day for 4 days, n = 10), (3) 5-fluorouracil + sham FMT group (5-fluorouracil intraperitoneal injection, and fed with PBS via oral gavage once a day for 4 days, n = 8), and (4) 5-fluorouracil + FMT group (5-fluorouracil intraperitoneal injection, and fed with the fecal suspension from normal rats via oral gavage once a day for 4 days, n = 8). Then, 1H nuclear magnetic resonance (NMR) spectroscopy-based metabolic phenotyping was used to investigate the changes of prefrontal cortex (PFC) metabolites affected by 5-fluorouracil.
