Stable isotope labeling derivatization and magnetic dispersive solid phase extraction coupled with UHPLC-MS/MS for the measurement of brain neurotransmitters in post-stroke depression rats administrated with gastrodin

Overall design

In this study, d0/d3-6-N-methyl-rhodamine 6G-N-hydroxysuccinimidyl formate (d0/d3-MRSF) were firstly designed, synthesized and used as stable isotope labeling derivatization reagents for the determination of multiple neurotransmitters by ultra-high performance liquid chromatography-tandem mass spectrometry. The developed method was applied to the analysis of endogenous neurotransmitters in brain and exogenous gastrodin in blood from the control and poststroke depression (PSD) model rats. Sprague-Dawley rats were divided into the following 2 groups (n = 6 in each group): (1) control group, and (2) PSD group (left middle cerebral artery occlusion with unpredictable chronic mild stress). The rats in PSD rats group experienced 3 stages with in vivo microdialysis state, stage A, putative PSD, 120 min; stage B, the rats were treated with gastrodin (200 mg/kg), about 20 min the blood concentration of gastrodin reached the peak, 220 min later gastrodin could not be detected in the blood microdialysates; after the effect of gastrodin disappeared for 120 min, rats entered into stage C with treated with fluoxetine (10 mg/kg) and received in vivo microdialysis sampling experiments for 300 min. The rats in the control group received an equivalent volume of saline.
