Deciphering the differential effective and toxic responses of Bupleuri Radix following the induction of chronic unpredictable mild stress and in healthy rats based on serum metabolic profiles

Overall design

This study aimed to decipher the differential effective and toxic responses of Bupleuri Radix (PBR) in chronic unpredictable mild stress (CUMS) and healthy rats based on serum metabolic profiles. Rats were divided into the following 17 groups (n = 8 in each group): (1) control group, (2) CUMS group, (3-9) seven different doses of PBR group (stressor plus Bupleuri Radix treatment at the dose of concentration of D1 - D7, respectively), (10) CUMS + venlafaxine group (stressor plus venlafaxine treatment at the dose of 25 mg/kg), and (11-17) CUMS + seven different doses of PBR group (stressor plus Bupleuri Radix treatment at the dose of concentration of D1 - D7). The serum samples of rats in the CUMS group, CUMS + D4 doses of PBR group (D4 dose of PBR), CUMS + D6 doses of PBR group (D4 dose of PBR), CUMS + D7 doses of PBR group (D4 dose of PBR), control group, D4 doses of PBR group (D4 dose of PBR), D6 doses of PBR group (D4 dose of PBR), D7 doses of PBR group (D4 dose of PBR) were used to obtain metabolic profiling data for metabolomics analysis using UHPLC- Q Exactive Orbitrap-MS technique. The CUMS stress procedure lasted for 3 weeks, and drugs were administered by gavage once per day during the model building period.
