Effects of refined Xiaoyaosan on depressive-like behaviors in rats with chronic unpredictable mild stress through neurosteroids, their synthesis and metabolic enzymes

Overall design

The aim of this study was to observe the effects of refined Xiaoyaosan (XYS) on the depressive-like behaviors in rats with chronic unpredictable mild stress (CUMS), and to explore the relationship between the changes of neurosteroids and mRNA expressions of their synthesis and metabolic enzymes, and the mechanism of XYS in the treatment of depression. The levels of pregnenolone, progesterone and alloprognanolone in the hippocampus and amygdala tissues were measured by LC-MS. All the rats were divided into the following 4 groups (n = 21 in each group): (1) control group, (2) CUMS group, (3) CUMS + XYS group (stress plus refined Xiaoyaosan treatment at the dose of 0.197 g/kg), and (4) CUMS + fluoxetine group (stress plus fluoxetine treatment at the dose of 2 mg/kg). Six samples per group were used for LC-MS analysis. The CUMS stress procedure lasted for 3 weeks, and drugs were administered via intragastric once per day during the model building period.
