Title | Serum metabonomics study of anti-depressive effect of Xiao-Chai-Hu-Tang on rat model of chronic unpredictable mild stress |
Overall design | In this study, the metabolic profiles and potential biomarkers were investigated in a chronic unpredictable mild stress (CUMS) model after treatment with Xiao-Chai-Hu-Tang (XCHT). Metabonomics based on ultra-high performance liquid chromatography coupled with mass spectrometry was used to profile the metabolic fingerprints of serum obtained from a rat model with chronic unpredictable mild stress with and without XCHT treatment. Rats were divided into 4 groups (n = 8 in each group): (1) control group, (2) CUMS group, (3) CUMS + fluoxetine group (stress plus fluoxetine treatment at the dose of 20 mg/kg), and (4) CUMS + XCHT group (stress plus XCHT treatment at the dose of 5 g/kg). The CUMS stress procedure lasted for 6 weeks, and drugs were intragastric administration once per day during the last 4 weeks of the model building period. |
