Integration of non-targeted metabolomics and targeted quantitative analysis to elucidate the synergistic antidepressant effect of Bupleurum chinense DC-Paeonia lactiflora Pall herb pair by regulating purine metabolism

Overall design

The aim of this study was to investigate the compatibility mechanism of Chaihu and Baishao for treating depression in the chronic unpredictable mild stress (CUMS) model of depression. Sprague-Dawley rats were divided into the following 4 groups: (1) control group, (2) CUMS group, (3) CUMS + Chaihu group (stressor plus Chaihu treatment at the dose of 7.5 crude drug/kg), and (4) CUMS + Chaihu-Baishao herb pair group (stressor plus Chaihu-Baishao herb pair treatment at the dose of 7.5 crude drug/kg). The CUMS stress procedure lasted for 4 weeks, and drugs were administered via intragastric once per day during the model building period. Cortex metabolomics based on ultra-high-performance liquid chromatography combined with quadrupole orbitrap mass spectrometry was used to discover the metabolic pathway that was synergistically regulated by Chaihu-Baishao herb pair. Based on the results of untargeted metabolomics analysis, metabolites were quantitatively validated by UPLC-MS/MS combined with the MRM mode in the crucial metabolic pathway. Six samples per group were used for both untargeted and targeted metabonomic analysis.
