Overall design | The aim of this study was to investigate the compatibility mechanism of Chaihu and Baishao for treating depression in the chronic unpredictable mild stress (CUMS) model of depression. Sprague-Dawley rats were divided into the following 4 groups: (1) control group, (2) CUMS group, (3) CUMS + Chaihu group (stressor plus Chaihu treatment at the dose of 7.5 crude drug/kg), and (4) CUMS + Chaihu-Baishao herb pair group (stressor plus Chaihu-Baishao herb pair treatment at the dose of 7.5 crude drug/kg). The CUMS stress procedure lasted for 4 weeks, and drugs were administered via intragastric once per day during the model building period. Cortex metabolomics based on ultra-high-performance liquid chromatography combined with quadrupole orbitrap mass spectrometry was used to discover the metabolic pathway that was synergistically regulated by Chaihu-Baishao herb pair. Based on the results of untargeted metabolomics analysis, metabolites were quantitatively validated by UPLC-MS/MS combined with the MRM mode in the crucial metabolic pathway. Six samples per group were used for both untargeted and targeted metabonomic analysis. |
