Title | Integrated network pharmacology and GC-MS-based metabolomics to investigate the effect of Xiang-Su volatile oil against menopausal depression |
Overall design | The aim of this study was to investigate the pharmacological mechanism of Xiang-Su volatile oil against menopausal depression through network pharmacology and metabolomics. Antidepressant-like effect of Xiang-Su volatile oil was validated in menopausal rats by ovariectomy combined with chronic unpredictable mild stress (CUMS). Sprague-Dawley rats were divided into the following 3 groups: (1) control group (pseudo-operation), (2) ovariectomy + CUMS group, and (3) ovariectomy + CUMS + Xiang-Su volatile oil group. Bilateral ovariectomy was performed in the rats (day 0), then rats received CUMS and Xiang-Su volatile oil (97.2 mg/kg, gastric perfusion once daily) from day 15 to day 28. GC-MS-based non-targeted plasma metabolomics were employed to validate the antidepressant effect of Xiang-Su volatile oil (n = 8-10/per group). |
