Urinary metabonomics study of anti-depressive mechanisms of Millettia speciosa Champ on rats with chronic unpredictable mild stress-induced depression

Overall design

In this study, urine metabolomics based on ultra-performance liquid chromatography/quadrupole time of flight mass spectrometry combined with pharmacodynamics was used to explore the pathogenesis of chronic unpredictable mild stress (CUMS)-induced depression and the antidepressant effects of Millettia speciosa Champ. Sprague-Dawley rats were divided into the following 6 groups (n = 6 per group): (1) control group, (2) CUMS group, (3) CUMS + high dose of Millettia speciosa Champ group (stressor plus Millettia speciosa Champ treatment at the dose of 14 g/kg), (4) CUMS + middle dose of Millettia speciosa Champ group (stressor plus Millettia speciosa Champ treatment at the dose of 7 g/kg), (5) CUMS + low dose of Millettia speciosa Champ group (stressor plus Millettia speciosa Champ treatment at the dose of 3.5 g/kg), and (6) CUMS + fluoxetine group (stressor plus fluoxetine treatment at the dose of 2.1 mg/kg). Urine samples (n = 5-6/group) were used for metabonomic analysis.
