Title

Gut microbiota and gut tissue metabolites involved in development and prevention of depression

Overall design

In this study, the authors investigated the metabolic signatures in the jejunum, ileum, and colorectum using metabolomics and explored the influence of the gut microbiota on both the development of chronic variable stress (CVS)-induced depression rat model and variations in gut tissue metabolites using a gnotobiotic rat model. Wistar rats were divided into the following 3 groups (n = 8 per group): (1) control group, (2) CVS group, and (3) CVS + antibiotic group. The CVS stress procedure lasted for 4 weeks, and antibiotics (imipenem/cilastatin sodium) was administered at a daily dose of 75 mg/kg of body weight from day 25 to day 28. Alteration in the metabolites in the jejunum, ileum, and colorectum of CVS rats were investigated using UPLC-Q-TOF/MS analysis. Influence of antibiotics on the differential metabolites in gut tissues related to depression were also explored.
