Title | Wenyang Jieyu decoction alleviates depressive behavior in rat model of depression via regulation of the metabolism of short-chain fatty acids in intestinal microbiota |
Overall design | The aim of this study was to investigate the effect of Wenyang Jieyu decoction (WYJYD) on rat model of depression based on metabolomics. The rat model of depression was established by chronic unpredictable mild stress (CUMS). Sprague-Dawley rats were divided into the following 5 groups (n = 6 in each group): (1) control group, (2) CUMS group, (3) CUMS + middle dose of WYJYD group (stressor plus WYJYD treatment at the dose of 3.08 g/kg/d), (4) CUMS + high dose of WYJYD group (stressor plus WYJYD treatment at the dose of 7.56 g/kg/d), and (5) CUMS + fluoxetine group (stressor plus fluoxetine treatment at the dose of 3.33 mg/kg/d. The CUMS stress procedure lasted for 4 weeks, and drugs were administered via intragastric once per day during the model building period. The short-chain fatty acid contents in intestinal contents were detected with GC-MS. |
