Title | A high coverage pseudotargeted lipidomics method based on three-phase liquid extraction and segment data-dependent acquisition using UHPLC-MS/MS with application to a study of depression rats |
Overall design | This study proposed a comprehensive pseudotargeted lipidomics method based on three-phase liquid extraction (3PLE) and segment data-dependent acquisition (SDDA). The established pseudotargeted method was applied to the plasma lipid profiling from the chronic unpredictable mild stress (CUMS) rat model of depression. Sprague-Dawley rats were divided into the following 4 groups (n = 8 in each group): (1) control group, (2) CUMS group, (3) CUMS + liquiritin group (stressor plus liquiritin suspension treatment at the dose of 20 mg/kg), and (4) CUMS + fluoxetine group (stressor plus fluoxetine treatment at the dose of 10 mg/kg). The CUMS stress procedure lasted for 4 weeks, and drugs were administered via intragastric once per day during the model building period. |
