Enhanced pseudotargeted analysis using a segment data dependent acquisition strategy by liquid chromatography-tandem mass spectrometry for a metabolomics study of liquiritin in the treatment of depression

Overall design

In this study, an enhanced pseudotargeted method using a segment data-dependent acquisition mode based on ultra-high performance liquid chromatography-tandem mass spectrometry was developed. The segment data dependent acquisition-based pseudotargeted method was successfully applied to the urine metabolomics study of the chronic unpredictable mild stress (CUMS) model of depression with the treatment of liquiritin. Sprague-Dawley rats were divided into the following 4 groups (n = 8 in each group): (1) control group, (2) CUMS group, (3) CUMS + liquiritin group (stressor plus liquiritin suspension treatment at the dose of 20 mg/kg), and (4) CUMS + fluoxetine group (stressor plus fluoxetine treatment at the dose of 10 mg/kg). The CUMS stress procedure lasted for 4 weeks, and drugs were administered via intragastric once per day during the model building period.
