Overall design | In this study, an integrated 1H NMR-based metabolomic approach was applied to screen and identify biomarkers associated with chronic unpredictable mild stress (CUMS) mice and understand the antidepressive influences of iridoid-rich fraction of V. jatamansi (IRFV) in a depressive mouse model. Kunming mice were divided into the following 5 groups (n = 7 in each group): (1) control group, (2) CUMS group, (3) CUMS + fluoxetine group (stressor plus fluoxetine treatment at the dose of 2.5 mg/kg), (4) CUMS + middle dose of iridoid-rich fraction of V. jatamansi group (stressor plus iridoid-rich fraction of V. jatamansi treatment at the dose of 11.47 mg/kg), and (5) CUMS + high dose of iridoid-rich fraction of V. jatamansi group (stressor plus iridoid-rich fraction of V. jatamansi treatment at the dose of 22.94 mg/kg). The CUMS stress procedure lasted for 4 weeks, and drugs were administered via intragastric once per day during the model building period. |
