Overall design | The aim of this study was to explores the antidepressant mechanism of Radix Bupleuri-Radix Paeoniae Alba herb pair and the core role in Xiaoyaosan. The behavioral indicators were used to observe the effects of herb pair, Xiaoyaosan, and a negative control (Xiaoyaosan remove Radix Bupleuri-Radix Paeoniae Alba) on chronic unpredictable mild stress (CUMS) rats. Sprague-Dawley rats were divided into the following 8 groups (n = 10 in each group): (1) control group, (2) CUMS group, (3) CUMS + venlafaxine group (stressor plus venlafaxine treatment at the dose of 35 mg/kg), (4) CUMS + high dose of Xiaoyaosan group (stressor plus Xiaoyaosan treatment at the dose of 46 g/kg), (5) CUMS + low dose of Xiaoyaosan group (stressor plus Xiaoyaosan treatment at the dose of 23 g/kg), (6) CUMS + high dose of Radix Bupleuri-Radix Paeoniae Alba herb pair group (stressor plus Radix Bupleuri-Radix Paeoniae Alba herb pair treatment at the dose of 15 g/kg), (7) CUMS + low dose of Radix Bupleuri-Radix Paeoniae Alba herb pair group (stressor plus Radix Bupleuri-Radix Paeoniae Alba herb pair treatment at the dose of 7.5 g/kg), and (8) CUMS + high dose of negative control treatment group (stressor plus Xiaoyaosan remove Radix Bupleuri-Radix Paeoniae Alba treatment at the dose of 31 g/kg). The CUMS stress procedure lasted for 4 weeks, and drugs were administered via intragastric once per day during the model building period. Seven serum samples per group were used for NMR-based metabonomic analysis. |
