[Active substance fractions and effects of Shugan Hewei Tang on hippocampal neurotransmitters in rat model of chronic stress-induced depression]

Overall design

The aim of this study was to investigate the effects of Shugan Hewei Decoction (SGHWT) and its active substance fractions (volatile oils, polysaccharides, and terpenoids) on behavior and neurotransmitter levels in hippocampus of chronic unpredictable mild stress (CUMS) depression model rats. Sprague-Dawley rats were divided into the following 11 groups (n = 8 in each group): (1) control group, (2) CUMS group, (3) CUMS + fluoxetine group (stressor plus fluoxetine treatment at the dose of 1.58 g/kg), (4) CUMS + low dose of SGHWT group (stressor plus SGHWT treatment at the dose of 3.67 g/kg), (5) CUMS + high dose of SGHWT group (stressor plus SGHWT treatment at the dose of 14.68 g/kg), (6) CUMS + low dose of volatile oils group (stressor plus volatile oils treatment at the dose of 0.018 g/kg), (7) CUMS + high dose of volatile oils group (stressor plus volatile oils treatment at the dose of 0.073 g/kg), (8) CUMS + low dose of polysaccharides group (stressor plus polysaccharides treatment at the dose of 0.56 g/kg), (9) CUMS + high dose of terpenoids group (stressor plus terpenoids treatment at the dose of 0.012 g/kg), (10) CUMS + low dose of alkaloids group (stressor plus alkaloids treatment at the dose of 0.013 g/kg), and (11) CUMS + high dose of phenolic acids group (stressor plus phenolic acids treatment at the dose of 0.13 g/kg). The CUMS stress procedure lasted for 3 weeks, and drugs were administered via intragastric once per day for 1 week after stress. Several neurotransmitters in hypothalamus of rats were determined by LC-MS/MS method, including norepinephrine, serotonin, 5-indoleacetic acid, GABA, and glutamic acid.
