Title

Elevated spectroscopic glutamate/gamma-amino butyric acid in rats bred for learned helplessness

Overall design

In this study, the effect of alterations of gamma-aminobutyric acid (GABA) and glutamate (Glu) were investigated in the congenital learned helplessness (cLH) paradigm. Proton magnetic resonance spectroscopy is an important monitoring tool to bridge the findings in clinical and preclinical studies. Rats were divided into the following 6 groups: (1) control group (outbred SD rats treated with saline, n = 8), (2) desipramine group (outbred SD rats treated with desipramine at the dose of 10 mg/kg, n = 10), (3) ECS group (outbred SD rats treated with electroconvulsive shocks, n = 8), (4) cLH group (cLH rats treated with saline, n = 5), (5) cLH + desipramine group (cLH rats treated with desipramine at the dose of 10 mg/kg, n = 5), and (6) cLH + ECS group (cLH rats treated with electroconvulsive shocks (ECS), n = 6). Desipramine was treated for 4 weeks with daily intraperitoneal injections. The ECS group received a 4-week course of ECS, administered three times per week.
