Overall design | The aim of this study was to investigate the hypothesis that Danzhi Xiaoyao San (DXS) treats depressive-like behavior by shifting the balance of the kynurenine (Kyn)/serotonin (5-HT) pathway toward the 5-HT pathway through the downregulation of hippocampal indoleamine 2,3-dioxygenase (IDO). Rats were divided into the following 4 groups (n = 10 in each group): (1) control group, (2) CUMS group, (3) CUMS + DXS group (stressor plus Danzhi Xiaoyao San treatment at the dose of 4 g/kg), and (4) CUMS + fluoxetine group (stressor plus fluoxetine treatment at the dose of 2 mg/kg). Cytokines, IDO, and tryptophan (Trp) catabolites were analyzed by enzyme-linked immunosorbent assay, western blot, and liquid chromatography-electrospray ionization tandem mass spectrometry, respectively. The CUMS stress procedure lasted for 3 weeks, and drugs were administered by gavage using a tube twice a day during the model building period. |
