Title | Dysfunctional glutamatergic and gamma-aminobutyric acidergic activities in prefrontal cortex of mice in social defeat model of depression |
Overall design | The aim of this study was to investigate molecular mechanisms in the prefrontal cortex of mice showing depression-like phenotype induced by chronic defeat stress. Depression-like phenotype was induced in C57BL/6 mice by subjecting them to a 10-day social defeat paradigm. The metabolic activity of excitatory (glutamatergic) and inhibitory (gamma-aminobutyric acid [GABA]ergic) neurons of the prefrontal cortex was measured by 1H-[13C]-nuclear magnetic resonance spectroscopy together with infusion of [1,6-13C2]glucose. Mice were divided into two groups: (1) control group (n = 15) and (2) SD group (social defeat paradigm; n = 30) for NMR and molecular studies. The social defeat paradigm procedure lasted for 10 days. Mice subjected to social defeat having an social interaction score less than 100 and reduced preference for sucrose were considered depressed and the rest were considered resilient. |
