Multiomics approach to explore the different amelioration mechanisms of Chaihu Shugan San and Hypericum perforatum L. on the metabolic disorder of CUMS-induced depressive rats

Overall design

The aim of this study was to explore potential antidepressant mechanisms of Chaihu Shugan San and Hypericum perforatum L. by analyzing ultra-performance liquid chromatography mass spectrometry metabolomics in chronic unpredictable mild stress (CUMS) rats. Sprague-Dawley rats were divided into the following 4 groups (n = 6 in each group): (1) control group, (2) CUMS group, (3) CUMS + Chaihu Shugan San group (stress with Chaihu Shugan San treatment at the dose of 19.8 g/kg), and (4) CUMS + Hypericum perforatum L. group (stress with Hypericum perforatum L. treatment at the dose of 204 mg/kg). The CUMS stress procedure lasted for 12 weeks, and drugs were administered via intragastric once per day during the last 8 weeks of the model building period. Serum samples were used for global metabonomic analysis.
