Title | Effects of Danggui-Shaoyao-San on central neuroendocrine and pharmacokinetics in female ovariectomized rats |
Overall design | The aim of this study was to explore whether the central oxytocin and neuroendocrine system is involved in the modulating effects of Danggui-Shaoyao-San on the cognition and neuropsychiatric hebaviors in female ovariectomized (OVX) rats. Female Sprague-Dawley rats were divided into the following 5 groups: (1) control group (sham ovariectomy with saline treatment), (2) ovariectomy group (ovariectomy with saline treatment), (3) ovariectomy + low dose of Danggui-Shaoyao-San group (ovariectomy with Danggui-Shaoyao-San treatment at the dose of 0.139 g/kg), (4) ovariectomy + middle dose of Danggui-Shaoyao-San group (ovariectomy with Danggui-Shaoyao-San treatment at the dose of 0.372 g/kg), and (5) ovariectomy + high dose of Danggui-Shaoyao-San group (ovariectomy with Danggui-Shaoyao-San treatment at the dose of 1 g/kg). After 15 days of operation, rats were administrated with Danggui-Shaoyao-San or saline via intragastric for 30 days. The concentrations of acetylcholine, glutamate, gamma-aminobutyric acid, 5-hydroxytryptamine, norepinephrine and dopamine in the hippocampus were assessed by HPLC-MS (n = 4 in each group). |
