Intranasal (R, S)-ketamine delivery induces sustained antidepressant effects associated with changes in cortical balance of excitatory/inhibitory synaptic activity

Overall design

The aim of this study was to compare the efficacy of a single intranasal dose of ketamine (10 and 20 mg/kg) versus a systemic intraperitoneal administration to promote a sustained antidepressant-like effect (at 24 h) and its consequences on the balance of excitation/inhibition synaptic transmission in the medial prefrontal cortex in hyperanxious BALB/cJ mice. BALB/cJ mice were divided into the following 4 groups: (1) intraperitoneal control group (vehicle treatment, intraperitoneally), (2) intraperitoneal ketamine group (ketamine treatment, 10 mg/kg, intraperitoneally), (3) intranasal control group (vehicle treatment, intranasally), and (4) intranasal ketamine group (ketamine treatment, 20 mg/kg, intranasally). (R, S)-ketamine (10 and 20 mg/kg) was administered intranasally or intraperitoneally in mice 24 h before the behavioral tests and in vivo microdialysis experiments. The extracellular levels of serotonin, glutamate, and GABA in the medial prefrontal cortex were measured by in vivo microdialysis experiments and LC-MS analysis (n = 12 to 17/group).
