Prophylactic effect of Tongxieyaofang polysaccharide on depressive behavior in adolescent male mice with chronic unpredictable stress through the microbiome-gut-brain axis

Overall design

The aim of this study was to evaluate the effect of Tongxieyaofang polysaccharide on intestinal flora metabolites in chronic unpredictable stress (CUS) model of depression. C57BL/6 mice were divided into the following 8 groups (n = 12 in each group): (1) control group, (2) CUS group, (3) CUS + low dose of Tongxieyaofang neutral polysaccharide group (stressor plus Tongxieyaofang neutral polysaccharide treatment at the dose of 50 mg/kg), (4) CUS + middle dose of Tongxieyaofang neutral polysaccharide group (stressor plus Tongxieyaofang neutral polysaccharide treatment at the dose of 100 mg/kg), (5) CUS + high dose of Tongxieyaofang neutral polysaccharide group (stressor plus Tongxieyaofang neutral polysaccharide treatment at the dose of 200 mg/kg), (6) CUS + Tongxieyaofang acidic polysaccharide group (stressor plus Tongxieyaofang acidic polysaccharide treatment at the dose of 100 mg/kg), (7) CUS + Tongxieyaofang neutral polysaccharide + Tongxieyaofang acidic polysaccharide group (stressor plus Tongxieyaofang neutral polysaccharide and Tongxieyaofang acidic polysaccharide treatment at the dose of 100 mg/kg), and (8) CUS + fructooligosaccharide + galactooligosaccharides group (stressor plus fructooligosaccharide and galactooligosaccharides treatment at the dose of 100 mg/kg). The CUS stress procedure lasted for 4 weeks, and drugs were administered via intragastric once per day during the model building period. Six fecal samples per group were used for metabonomic analysis.
