Overall design | The objective of this study is to compare the antidepressive effects of Xiaoyao Powder (XYP) composed of Bupleurum scorzonerifolium (Nan Chaihu) or B. chinense (Bei Chaihu) and to investigate their effects on the changes of endogenous metabolites in rats. The ethology symptoms of CUMS rats were observed and 1H-NMR method was carried out to analyze the changes of metabolites in serum of rats after drug administration. Rats were divided into five groups (n = 12 in each group): (1) control group, (2) CUMS group, (3) CUMS + Xiaoyaosan Nan group (stress plus Xiaoyaosan Nan at the dose of 46 g/kg), (4) CUMS + Xiaoyaosan Bei group (stress plus Xiaoyaosan Bei at the dose of 46 g/kg), and (5) CUMS + venlafaxine group (stress plus venlafaxine at the dose of 0.05 g/kg). Eight samples per group were used for metabonomic analysis. The CUMS stress procedure lasted for 4 weeks, and drugs were administered i.g. once a day during the last 2 weeks of model building period. |
