Title

Antidepressant-like effects of the fractions of Xiaoyaosan on rat model of chronic unpredictable mild stress

Overall design

The aim of this study was to evaluate the antidepressant activity of five different polar fractions of XYS to identify the active fraction using behavior research and metabonomics method-based on NMR. Behavior research and metabonomics method based on 1H NMR were used for efficacy study of different fractions in chronic unpredictable mild stress (CUMS) model of depression. The urine samples of rats were collected overnight on 21 day and the metabolic profiling of the urine was measured using NMR. Rats were randomly divided into 5 groups (n = 8 in each group): (1) control group, (2) CUMS group, (3) CUMS + XY-A group (stress plus XY-A treatment at the dose of 0.24 g/kg), (4) CUMS + XY-D group (stress plus XY-D treatment at the dose of 0.23 g/kg), and (5) CUMS + Xiaoyaosan group (stress plus Xiaoyaosan treatment at the dose of 5.98 g/kg). Six samples per group were used for metabonomic analysis. The CUMS stress procedure lasted for 3 weeks, and drugs were administered by gastric intubation one time each day during the model building period.
