[Effects of Jiaotai Pills on CUMS-induced depression model in mice based on changes of SIRT1 expression in hippocampus]

Overall design

The aim of this study was to investigate the effects and mechanisms of Jiaotai Pills on depressed mice induced by chronic unpredictable mild stress (CUMS). BALB/c mice were divided into the following 5 groups (n = 12 in each group): (1) control group, (2) CUMS group, (3) CUMS + fluoxetine group (stressor plus fluoxetine treatment at the dose of 3 mg/kg), (4) CUMS + low dose of Jiaotai Pills group (stressor plus Jiaotai Pills treatment at the dose of 2.5 g/kg), and (5) CUMS + high dose of Jiaotai Pills group (stressor plus Jiaotai Pills treatment at the dose of 5 g/kg). The CUMS stress procedure lasted for 3 weeks, and drugs were administered via intragastric once per day for 2 weeks after stress. Liquid chromatography-tandem mass spectrometry was used to detect the levels of dopamine, 5-hydroxytryptamine, and norepinephrine in the hippocampus (n = 6/per group) and serum (n = 12/per group) of mice.
