Integrated network pharmacology analysis and experimental validation to investigate the mechanism of Zhi-Zi-Hou-Po Decoction in depression

Overall design

The aim of this study was to explore the pharmacological mechanisms of Zhi-Zi-Hou-Po Decoction acting on depression by combining ultra flow liquid chromatography with quadrupole time-of-flight mass spectrometry and network pharmacology strategy. Sprague-Dawley rats were divided into the following 5 groups (n = 8 in each group): (1) control group, (2) corticosterone group, (3) corticosterone + middle dose of Zhi-Zi-Hou-Po Decoction group (corticosterone plus Zhi-Zi-Hou-Po Decoction treatment at the dose of 6 g/kg), (4) corticosterone + high dose of Zhi-Zi-Hou-Po Decoction group (corticosterone plus Zhi-Zi-Hou-Po Decoction treatment at the dose of 12 g/kg), and (5) corticosterone + fluoxetine group (corticosterone plus fluoxetine treatment at the dose of 20 mg/kg). Corticosterone was administered subcutaneously to induce depression at 20 mg/kg once daily for 21 days. Zhi-Zi-Hou-Po Decoction and fluoxetine were orally administrated each day 30 min prior to the corticosterone injection. The levels of serotonin and dopamine in the hippocampal samples were analyzed by LC-MS/MS.
