Baihe Jizihuang Tang ameliorates chronic unpredictable mild stress-induced depression-like behavior: integrating network pharmacology and brain-gut axis evaluation

Overall design

The aim of this study was to clarify the antidepressive effects and molecular mechanism of Baihe Jizihuang Tang in the chronic unpredictable mild stress (CUMS) model of depression. Sprague-Dawley rats were divided into the following 5 groups: (1) control group, (2) CUMS group, (3) CUMS + fluoxetine group (stressor plus fluoxetine treatment at the dose of 1.8 mg/kg), (4) CUMS + middle dose of Baihe Jizihuang Tang group (stressor plus Baihe Jizihuang Tang treatment at the dose of 5.4 g/kg), and (5) CUMS + high dose of Baihe Jizihuang Tang group (stressor plus Baihe Jizihuang Tang treatment at the dose of 10.8 g/kg). The CUMS stress procedure lasted for 30 days, and drugs were administered via intragastric once per day during the model building period. Dopamine, norepinephrine, serotonin, and glutamic acid concentrations in the hippocampus were determined by UPLC-MS. Six samples per group were used for targeted metabonomic analysis.
