Stimulation of peroxisome proliferator-activated receptor-alpha by N-palmitoylethanolamine engages allopregnanolone biosynthesis to modulate emotional behavior

Overall design

The aim of this study was to investigate a possible role for PPAR-a agonists in the upregulation of allopregnanolone synthesis as a means for reducing PTSD- and depression- or anxiety-like behaviors in socially isolated mice with allopregnanolone deficits. Socially isolated mice were housed individually for 5 weeks, while group-housed mice were housed in groups of 5. Socially isolated mice showed depression- or anxiety-like behaviors compared with group-housed mice. Median effective concentration dose of N-palmitoylethanolamine, given to socially isolated mice 1 hour before behavior tests, significantly decreased immobility in the forced swim test and tail suspension test. The sulfated neurosteroids pregnenolone sulfate, allopregnanolone sulfate, pregnanolone sulfate, and dehydroepiandrosterone sulfate were determined by gas chromatography-mass spectrometry (n = 6 -7).
