Title

Substance P receptor antagonist and clomipramine prevent stress-induced alterations in cerebral metabolites, cytogenesis in the dentate gyrus and hippocampal volume

Overall design

The present study investigated the effect of the NK1R antagonist L-760,735 and the tricyclic antidepressant clomipramine in the chronic psychosocial stress paradigm of adult male tree shrews. Animals were divided into the following 4 groups: (1) control group, (2) stress group (daily psychosocial conflict stress), (3) stress + L-760,735 group (daily psychosocial conflict stress plus L-760,735 treatment at the dose of 10 mg/kg), and (4) stress + clomipramine group (daily psychosocial conflict stress plus clomipramine treatment at the dose of 50 mg/kg). The psychosocial conflict stress procedure lasted for 5 weeks, and drugs were administered via tap water during the last 4 weeks of the model building period. Brain metabolite concentrations were determined in vivo by proton magnetic resonance spectroscopy. Sample sizes were 9 for control group, 11 for stress group, 5 for stress + clomipramine group and 5 for stress + L-760,735 group. Metabolite quantification was reported relative to brain water concentration.
