Title

SONU20176289, a compound combining partial dopamine D(2) receptor agonism with specific serotonin reuptake inhibitor activity, affects neuroplasticity in an animal model for depression

Overall design

The aim of this study was to investigated the efficacy of SONU20176289, a member of a group of novel phenylpiperazine derivatives with a mixed dopamine D2 receptor partial agonist and specific serotonin reuptake inhibitor (SSRI) activity, in a chronic stress model of depression in male tree shrews. Animals were divided into the following 4 groups (n = 6 in each group): (1) control group, (2) SONU group (no stress, SONU20176289 treatment at the dose of 6 mg/kg), (3) stress group (daily psychosocial conflict stress), (4) stress + SONU group (daily psychosocial conflict stress plus SONU20176289 treatment at the dose of 6 mg/kg). The psychosocial conflict stress procedure lasted for 5 weeks, and drugs were administered orally during the last 4 weeks of the model building period. Urinary noradrenaline was quantified by LC-MS. Localized proton magnetic resonance spectroscopy was used to measure the in vivo brain concentrations. All animals underwent imagecontrolled localized proton MRS to obtain in vivo concentrations of major brain metabolites. Additional MRS data for animals of the control and the stress group were available from a cohort of age-matched males acquired in multiple studies. Metabolite quantification was reported relative to brain water concentration.
