Involvement of the glutamate/glutamine cycle and glutamate transporter GLT-1 in antidepressant-like effects of Xiao Yao san on chronically stressed mice

Overall design

This study aimed to assess the chronic antidepressant effects of Xiao Yao san (XYS) and fluoxetine in depressed mice induced by chronic unpredictable mild stress (CUMS) and its association with alterations in glutamate/glutamine cycle and glutamate transporters. Mice were divided into the following 4 groups: (1) control group, (2) CUMS group, (3) CUMS + XYS group (stressor plus Xiao Yao san treatment at the dose of 0.25 g/kg), and (4) CUMS + fluoxetine group (stressor plus fluoxetine treatment at the dose of 2.6 mg/kg). The CUMS stress procedure lasted for 3 weeks, and drugs were intragastrically administered once per day during the model building period. The bilateral hippocampus of five mice in each group were used for LC-MS analysis. The levels of glutamate (Glu) and glutamine (Gln) in the hippocampus were measured by LC-MS/MS.
