Regional alterations in the endocannabinoid system in an animal model of depression: effects of concurrent antidepressant treatment

Overall design

The aim of this study was to analysis the endocannabinoid system in an animal model of depression. For behavioral testing, one cohort of animals was either subjected to 21 days of chronic unpredictable stress (CUS) or acted as cage controls and were handled weekly. For biochemical assays, rats were divided into the following 4 groups (n = 8 in each group): (1) control group, (2) imipramine group (imipramine treatment at the dose of 10 mg/kg), (3) CUS group, (4) CUS + imipramine group (stressor plus imipramine treatment at the dose of 10 mg/kg). For LC-MS analysis, 7-8 samples per group were used. The CUS stress procedure lasted for 3 weeks, and imipramine was administered via intraperitoneal injection once per day during the model building period. The contents of the two primary endocannabinoids anandamide (AEA) and 2-arachidonoylglycerol (2-AG) within lipid extracts in methanol from brain tissue and plasma were determined using isotope-dilution liquid chromatography-mass spectrometry.
