Antidepressant-like effects and cognitive enhancement of the total phenols extract of Hemerocallis citrina Baroni in chronic unpredictable mild stress rats and its related mechanism

Overall design

The authors designed to evaluate the antidepressant and cognitive improvement effect of total phenols extract of Hemerocallis citrina Baroni (HCPE) using a chronic unpredictable mild stress (CUMS) model, and the potential mechanisms were explored by investigating the corticosterone (CORT), monoamine neurotansmitters, brain-derived neurotropic factor (BDNF) and oxidative stress. The depression rats were induced by CUMS procedures and treated with HCPE (10, 20, 40 mg/kg/day, by gastric gavage). The antidepressant effect was evaluated by sucrose preference test, open field test and body weight, while the cognitive improvement was investigated using morris water maze test. Besides, the levels of monoamine neurotransmitters in the hippocampus and frontal cortex were measured by ultra-performance liquid chromatography tandem mass spectrometry (UPLC-MS/MS). Rats were divided into the following 6 groups (n = 10 in each group): (1) control group, (2) CUMS group, (3) CUMS + paroxetine group (stressor plus paroxetine treatment at the dose of 10 mg/kg), (4) CUMS + low dose of HCPE group (stressor plus HCPE treatment at the dose of 10 mg/kg), (5) CUMS + middle dose of HCPE group (stressor plus HCPE treatment at the dose of 20 mg/kg), and (6) CUMS + high dose of HCPE group (stressor plus HCPE treatment at the dose of 40 mg/kg). The CUMS stress procedure lasted for 5 weeks, and drugs were administered by gastric gavages once per day during the model building period.
