Title | Role of prefrontal cortical calcium independent phospholipase A2 in antidepressant-like effect of maprotiline |
Overall design | This study was conducted to elucidate a possible role of PFC activity of an enzyme involved in the release of docosahexaenoic acid (DHA), i.e. calcium-independent phospholipase A2 (iPLA2), in the effects of the norepinephrine reuptake inhibitor (NRI) antidepressant, maprotiline, in mice. Treatment of Balb/C mice with maprotiline for 4 week resulted in reduction in the level of behavioural despair, as determined by decreased immobility and increased climbing during the forced swim test. Lipid profiles of the prefrontal cortex were analyzed by high-performance liquid chromatography/mass spectrometry (HPLC/MS). The mice were divided into two equal groups (27 mice per group), and given daily i.p. injections of maprotiline (10 mg/kg in saline) or saline for 28 d. At day 21 of i.p. saline/maprotiline treatment, each group was further subdivided into three groups of 9 mice each. They were administered antisense oligonucleotide to iPLA2 (maprotiline + andantisense group and andantisense group), scrambled sense (maprotiline + sense group and sense group) or saline (maprotiline group and control group) via i.c. injection as described above and the FST was conducted 1 week later. Only data from maprotiline group (maprotiline treatment for 28 d) and control group (saline treatment for 28 d) were used. |
