Gut-microbiome-expressed 3beta-hydroxysteroid dehydrogenase degrades estradiol and is linked to depression in premenopausal females

Overall design

The aim of this study was to determine whether K. aerogenes TS2020 can induce depression-like behaviors in female mice. K. aerogenes-recipient mice was administered with cefotaxime to investigate whether serum estradiol reduction and depression-like behavior in the mice could be alleviated by antibiotic treatment. C57BL/6J mice were divided into the following 3 groups: (1) ovariectomy + estradiol group (ovariectomy followed by gavage estradiol for 6 weeks), (2) ovariectomy + estradiol + K. aerogenes TS2020 group (ovariectomy followed by gavage estradiol for 6 weeks and K. aerogenes TS2020 for 4 weeks), and (3) ovariectomy + estradiol + K. aerogenes TS2020 + cefotaxime group (ovariectomy followed by gavage estradiol for 6 weeks and K. aerogenes TS2020 for 4 weeks, then treated with cefotaxime for 1 week). The results of behavioral assessments showed that K. aerogenes TS2020 can induce depressive-like behaviors, and cefotaxime administration can alleviate such depressive-like behaviors in mice. Liquid chromatography-tandem mass spectrometry was used to compare serum estradiol levels between groups (n = 6/group).
