Title | NMR-based metabonomic analysis of normal rat urine and faeces in response to (+-)-venlafaxine treatment |
Overall design | With a pharmacometabonomic approach using NMR in combination with different chemometric methods, a determination was made of subtle changes in the metabolic profile (metabotype) of urine and fasces in normal Wistar rats following a single administration of pharmacological doses of (+-)-venlafaxine hydrochloride. All the rats were divided into 3 groups (n = 18 in each group): (1) control group (single dose of vehicle 5% Tween-80 w/v), (2) low dose of venlafaxine group (22 mg/kg of b.w. of venlafaxine free base, dose volume 4 mL/kg b.w.), and (3) high dose of venlafaxine group (112 mg/kg of b.w. of venlafaxine free base, dose volume 4 mL/kg b.w.). Urine samples were collected at intervals of 0-6 h, 6-12 h and 12-24 h. The collection period of stool samples was 0-24 h. |
