Title | [1H NMR based metabonomics study on the antidepressant effect of genipin in rat hippocampus] |
Overall design | The purpose of this study was to explore depression metabolic markers in rat hippocampus and to investigate the anti-depressant effect of genipin and its mechanisms using nuclear magnetic resonance (NMR) metabonomics. Chronic unpredictable mild stress (CUMS) procedure was conducted to establish the depressive rat model. Rats were divided into 6 groups (n = 8 in each group): (1) control group, (2) CUMS group, (3) CUMS + low dose of genipin group (stress plus genipin treatment at the dose of 25 mg/kg), (4) CUMS + middle dose of genipin group (stress plus genipin treatment at the dose of 50 mg/kg), (5) CUMS + high dose of genipin group (stress plus genipin treatment at the dose of 100 mg/kg), and (6) CUMS + venlafaxine group (stress plus venlafaxine treatment at the dose of 50 mg/kg). Rat hippocampus was analyzed by 1H NMR based metabonomics after drug administration for 2 weeks. The CUMS stress procedure lasted for 4 weeks, and drugs were administered via intragastric once per day during the last 2 weeks of model building period. |
